Background:The treatment of erectile dysfunction is generally thought to be a process of balancing certain components of the cardiovascular system and the vascular system, which is thought to be an underlying process that is not well understood. We hypothesized that patients with erectile dysfunction could benefit from sildenafil citrate (Viagra), a phosphodiesterase type 5 (PDE5) inhibitor. We performed this study to evaluate the safety and efficacy of sildenafil citrate in patients with erectile dysfunction.
Methods:We evaluated the safety of sildenafil citrate and dapoxetine for treatment of erectile dysfunction in a multicenter, double-blind, randomized, placebo-controlled clinical trial in 40 men with erectile dysfunction treated with sildenafil citrate and dapoxetine for 6 months. The study protocol was approved by the institutional review boards and was conducted in accordance with the Declaration of Helsinki and Good Clinical Practice guidelines.
Results:We found that patients with erectile dysfunction were more likely to report adverse events (AEs) compared to those not taking sildenafil citrate. Adverse events included the headache, flushing, dizziness, nasal congestion, and dyspepsia, and the erectile dysfunction was the most frequently reported AE. Sildenafil citrate, in combination with dapoxetine, was the most effective drug for the treatment of erectile dysfunction. The most common AEs reported with the combination of sildenafil citrate and dapoxetine were headache, dyspepsia, and nausea. The most common adverse events were headache, flushing, dyspepsia, dizziness, and nasal congestion. The most common AEs in both groups were hypotension, and erectile dysfunction was the most common of all AEs reported.
Conclusion:The safety profile of sildenafil citrate was similar to that of dapoxetine. Patients with erectile dysfunction were more likely to report AEs, including headache, flushing, dizziness, nasal congestion, and dyspepsia. The combination of sildenafil citrate and dapoxetine may be more effective than dapoxetine alone in the treatment of erectile dysfunction.
